Metabolism of GalNAc-siRNA conjugates is mainly catalyzed by exonucleases and endonucleases, resulting in various sizes of shortened oligonucleotides, which could interfere the quantitation of GalNAc-siRNA conjugates. Therefore, bioanalytical scientists have to use a long injection time to separate analyte from metabolites when using LC-MS/MS for quantitation.
